Stacking Tiers

I'm making my first wedding cake and I want to know:
If I use cake boards do I have to stack the cake (three tiers) first then put the dowels all the way through, or can it do one tier at a time?

If you use cake boards under each tier just put about 5 straws(cut to the height of the tier) in the center of the tier, in a little circle before putting the next tier on top. I read this in a cake decorating book and have used straws ever since. I have made 26 wedding cakes and this works great. It is cheaper than dowels and the "Wendys" size straws are great.
